P2X7, also known as P2Z or NLRP3, is a member of the purinergic receptor family of ligand-gated ion channels that includes P2X1-6 receptors. The immune system uses purines as signaling molecules, and ATP is the principal molecule of the purinergic signaling system. P2X7 is found predominantly on the cell surface of myeloid cells including macrophages and microglia, and contributes to cell death in neurodegenerative diseases. P2X7 functions to rapidly release cellular contents by forming a transmembrane pore following stimulation with extracellular ATP, triggering the assembly of the inflammasome complex, a multiprotein cytoplasmic complex required for pro-IL-1β cleavage to the mature form and subsequent release of IL-1β. P2X7 is also a key regulator of macrophage phagocytosis and inflammation through interactions with toll-like receptors (TLRs) and NLRs. P2X7 and TLR-NLR signaling has been shown to impact HIV replication and immune function, including macrophage-mediated killing of virus.
A potent and selective P2X7 receptor antagonist is a novel compound that, depending on its mode of administration, may be useful for treating or preventing diseases that have an inflammatory component. Specifically, such diseases may include pain, particularly neuropathic pain; chronic inflammatory diseases, such as, for example, asthma, emphysema, and rheumatoid arthritis; inflammatory bowel disease; allergy; and multiple sclerosis; non-inflammatory diseases such as, for example, diabetes, stroke, and muscle degeneration; and diseases such as cancer, cardiovascular disease, or neurodegenerative disease. The Council of the Cowlitz Indian Tribe of Washington was established by the Department of Interior in 1944 as part of the Indian Reorganization Act. It was a tribal council of the Cowlitz Indians of Washington. In 1951 it was revived as part of the termination of the Cowlitz Indian Reservation by the Department of Interior and was formally dissolved in 1968.
